TABLE 2-13:

Protease Inhibitor (PI) Resistance Mutations

Mutation Comment
L10I/V/F/R/V Accessory mutations that can contribute to reduced susceptibility in
the presence of other PI mutations. L10V: Contributes to TPV
resistance in presence of other TPV mutations.
V11I: Contributes to DRV resistance in presence of other DRV mutations
I13V Contributes to TPV resistance in presence of other TPV mutations.
K20R/I/M/T/V Accessory mutations/polymorphisms that may contribute to reduced
susceptibility in the presence of other PI mutations. K20M/R/V:
Contribute to TPV resistance in presence of other TPV mutations.
L23I Uncommon mutation; low-level NFV resistance.
L24I/F L24I: PI resistance, especially to IDV, when combined with other PI
mutations. L24F: Rare mutation with unknown effect on PI
susceptibility.
D30N Primary PI mutation selected only by NFV, especially with subtype B
virus; intermediate NFV resistance; further loss of susceptibility with
N88D/S.
V32I Accessory mutation that confers low-level resistance to IDV, RTV, APV,
LPV. Contributes to DRV resistance in presence of other DRV
mutations.
L33F/I/V L33F: decreases susceptibility to RTV, APV, LPV, ATV, TPV, and DRV
in presence of other PI mutations. L33I/V: Polymorphisms not known
to be associated with drug resistance.
E35G Contributes to TPV resistance in presence of other TPV mutations.
M36I/V/L M36I/V: Accessory mutations that contribute to reduced PI
susceptibility in the presence of other PI mutations. M36I: Contributes
to TPV resistance in presence of other TPV mutations. M36L:
Unknown effect on PI susceptibility.
K43T Contributes to TPV resistance in presence of other TPV mutations.
M46I/L/V M46I/L: Accessory mutation that contributes to reduced PI
susceptibility in the presence of other PI mutations. M46L: Contributes
to TPV resistance in presence of other TPV mutations. M46V:
Uncommon mutation with unknown effect on PI susceptibility.
I47A/V I47V: Reduced susceptibility to APV, IDV, RTV, LPV, TPV, and DRV in
combination with other PI mutations. I47A: Moderate to high-level LPV
resistance.
G48V/M G48V: selected by SQV; intermediate SQV resistance; low-level
resistance to other PIs. G48M: Effect on PI susceptibility unknown.
I50V/L I50V: Selected by APV in PI-naïve patients; intermediate APV
resistance; low to intermediate resistance to RTV, LPV. Contributes to
DRV resistance in presence of other DRV mutations. I50L: Selected by
ATV in PI-naïve patients; moderate to high-level ATV resistance;
susceptibility to other PIs maintained or increased.
F53L Associated with PI resistance when combined with other PI mutations.
